Planning a New 503B Facility? 5 Filling Equipment Decisions to Make Before Requesting a Quote
A new 503B outsourcing facility eventually reaches a point where planning turns into purchasing.
The production team has discussed the products.
The facility layout is taking shape.
The container supplier has been identified.
Then procurement sends the first email:
“Please send us your quotation for an aseptic filling machine.”
That sounds straightforward.
But there is a problem.
If the equipment requirements are not clearly defined, different suppliers may quote completely different machine configurations.
One supplier may quote a compact semi-automatic system.
Another may propose a fully automated line.
A third may recommend a multi-format platform.
Now the purchasing team has three prices — but no easy way to determine which system actually fits the project.
Before requesting a filling equipment quotation, five decisions can make the purchasing process much more efficient.
1. Decide Which Container Formats You Actually Need
This should be one of the first questions in the RFQ.
Are you filling:
Vials?
Prefilled syringes?
Cartridges?
Or potentially more than one format?
A facility that only plans to fill one vial size may have very different equipment requirements from a facility expecting several container formats.
For example, a new facility may initially plan:
2R and 10R vials
but already be discussing future cartridge-based products.
That information matters.
If the supplier knows the future requirements, the equipment configuration can be evaluated differently from a machine designed exclusively for the first production format.
Before requesting a quote, prepare:

- RTU or bulk

Don't quote only today's production plan if your facility is already planning tomorrow's products.
2. Define Your Fill Volume and Batch Size
The second decision is surprisingly important.
A machine designed around high-volume production may not be the most practical choice for a facility running smaller batches with frequent product changes.
Suppose a new outsourcing facility expects its initial production to be relatively small.
The team may value:

- Low product loss
- Flexibility
- Fast changeover
- Compact footprint
more than maximum theoretical output.
On the other hand, if the facility already has substantial production commitments, throughput and automation may become much more important.
So instead of telling a supplier:
“We need a pharmaceutical filling machine.”
provide:
Fill volume: 0.5 mL / 1 mL / 2 mL / etc.
Batch size: approximate number of containers
Target output: units/hour or units/batch
Production frequency: occasional / weekly / daily
These numbers immediately make the quotation more meaningful.
3. Decide How Much Automation You Really Need
This is where many equipment discussions become unnecessarily complicated.
A new facility may assume:
“We are building a professional facility, so we should buy a fully automatic filling line.”
Not necessarily.
The better question is:
“What level of automation matches our production volume and operating plan?”
Semi-Automatic
Can be attractive for:
- Smaller batches
- Clinical production
- Frequent product changes
- Flexible production
- Facilities scaling gradually
Automatic
Can become more attractive when:
- Production volume increases
- Labor requirements become significant
- Consistent throughput is important
- Production schedules become more predictable
- The facility needs higher automation
There is no universal “best” configuration.
The right choice depends on the relationship between:
Production Volume + Labor + Changeover + Future Growth
4. Think About Your Next Product Before Buying Your First Machine
This is where an equipment purchase can either become a long-term asset or a limitation.
Imagine your facility starts with vial filling.
Six months later, a customer asks about cartridge-based production.
The team goes back to the original machine supplier.
The question becomes:
“Can our existing filling system handle cartridges?”
If the answer is no, the facility may need another equipment investment.
This doesn't mean every new 503B facility should automatically purchase a multi-format machine.
It means future requirements should be discussed before the first purchase.
Ask your equipment supplier:
- Can the machine support multiple formats?
- What changeover is required?
- Which components need to be changed?
- Can future formats be added?
- What is the practical production range?
- Can the equipment be expanded later?
For a growing facility, these questions can be more valuable than comparing two machine prices.
5. Ask What Is Included in the Equipment Proposal

“Please quote your best filling machine.”
What Happens After You Send the RFQ?
A good equipment discussion should not end with a PDF quotation.
There should be a technical conversation around the project.

“What is your fill volume and batch size?”
Customer:
“2 mL, approximately 8,000 units per batch.”
Supplier:
“Are you planning only vials, or do you expect syringe or cartridge production later?”

Now the equipment discussion has changed.
